When using guide coat on filler and you find some low spots that need more filler. What is the method to use before applying filler to those low area with guide coat in them.......remove remaining guide coat with spi 700 or 710......or sand out those areas. ...or just apply the filler over the guide coat?

Just blow it off imho. That’s what I do.

Okay so the staining of the 3M dry coat after blowing it off has no effect on the next round for filler adhesion, that's good to know.

180 grit scratches okay to lay filler on?........or should it be scratched with 80 grit?

I usually blow it off with a red scotchbrite in the other hand. 180 is fine.

Perfect, I was removing the excess 3M guide coat with 180 grit sandpaper but I'll start using a scotchbrite, and thank you for answering the sand paper grit to use for application of filler on filler.

I remove any potential contamination with a red scuff pad, the only exception being if any area of filler is so low as to be untouched by sandpaper, that might need some extra attention with a coarser grit, then return with the red pad.
